ORLANDO, FL (UroToday.com) - The authors demonstrate that the widely employed chemotherapeutic agent Cisplatin (CP) based chemotherapy effects TSPY gene activity. A reverse transcription-polymerase chain reaction (RT-PCR) was employed to determine the expression of mRNA for TSPY and immunohistochemical staining for histone 2B (H2B) were performed on the rat testes of animals exposed to CP. There appeared to be a dose related effect on the expression of TSPY mRNA levels. Immunohistochemical analysis revealed that H2B expression was decreased in a dose dependent manner as a result of the destruction of DNA and chromosomes after exposure to CP.

Editorial Comment: This would support the notion that the infertility resulting from CISPLATINUM chemotheraoy may at least be partially explained in the basis of genetic alterations on the Y chromosome.
