Humulin R is a brand-name medication that’s available both over the counter and by prescription. It’s used to help manage blood sugar levels in adults and children with either type 1 or type 2 diabetes.
Humulin R comes as a liquid solution. It’s given as an injection under your skin (called a subcutaneous injection). You’ll give the injections to yourself once your doctor has shown you how to administer the medication.

Form of insulin in Humulin R
Humulin R contains the active drug insulin human. It belongs to a class of medications called short-acting insulins. (A medication class is a group of medications that work in a similar way). Humulin R is typically given two to three times each day before meals.
Insulin human is a “recombinant” insulin. This means it’s made in a lab using special DNA technology that combines genetic material in order to make the insulin. This process allows the drug manufacturer to create an exact copy of the insulin that your body makes naturally.
Effectiveness of Humulin R
In clinical trials, Humulin R was effective in improving blood sugar levels in people with either type 1 or type 2 diabetes. For information about the drug’s effectiveness, see the “Humulin R for diabetes” section below.
Humulin R is available only as a brand-name medication. It’s not currently available in generic form. A generic drug is an exact copy of the active drug in a brand-name medication.
Humulin R contains the active drug insulin human. The American Diabetes Association (ADA) also refers to this drug as “regular insulin,” “human insulin,” and “human regular insulin.” But no matter which name is used, they’re all referring to the same drug.
There is a drug available, called Novolin R, that’s similar to Humulin R. It contains the drug human insulin. For more information about this drug, see the section “Humulin R vs. Novolin R” below.
The Humulin R dosage your doctor prescribes will depend on several factors. These include:
- the type and severity of the condition you’re using Humulin R to treat
- your age
- the form of Humulin R you’re taking
- other medical conditions you may have
Typically, your doctor will start you on a low dosage. Then they’ll adjust it over time to reach the amount that’s right for you. Your doctor will ultimately prescribe the smallest dosage that provides the desired effect.
The following information describes dosages that are commonly used or recommended. However, be sure to take the dosage your doctor prescribes for you. Your doctor will determine the best dosage to fit your needs.
Note: Making changes to your insulin treatment plan without consulting your doctor could increase your risk for both hypoglycemia (low blood sugar levels) and hyperglycemia (high blood sugar levels). These changes may include administering your insulin differently than you usually do or injecting the drug into areas of skin that are thickened or pitted. They could also include taking a new dosage of insulin or using a new insulin product.
If your doctor changes your Humulin R dosage, they’ll likely have you monitor your blood sugar levels more often than usual. This allows you to monitor for high and low blood sugar levels.

Both the vials of Humulin R and the KwikPens are meant to be used more than once. However, the actual number of times you’ll use a certain vial or pen will depend on your insulin dosage.
But keep in mind that you’ll need to use a new syringe and a new needle for each dose of Humulin R from a vial. And you’ll need to use a new needle for each dose of insulin from a KwikPen. Using needles or syringes more than once can increase your risk of infection.

Dosage for diabetes
Usual dosages of Humulin R for both type 1 and type 2 diabetes are described below. However, your doctor will recommend a specific dosage that’s right for you.
If you have either type 1 or type 2 diabetes, your doctor will determine your insulin dosage based on your blood sugar levels and your blood sugar level goals.
For type 1 diabetes
The package information for Humulin R doesn’t contain exact dosage guidance for type 1 diabetes. But the American Diabetes Association (ADA) provides the following recommendations.
A typical starting dosage for type 1 diabetes is about 0.5 units of Humulin R per kilogram of your body weight per day (1 kilogram is equal to about 2.2 pounds). This daily dosage is split into two or three injections, which are each given about 30 minutes before your meals.
For type 2 diabetes
The package information for Humulin R doesn’t contain exact dosage guidance for type 2 diabetes. But the ADA provides the following recommendations.
A typical starting dosage for type 2 diabetes is around 0.1 to 0.2 units of Humulin R per kilogram of your body weight per day. (One kilogram is equal to about 2.2 pounds.) This daily dosage is split into two or three injections, which are each given about 30 minutes before your meals.
Sliding scale dosage
It’s also possible to use sliding scale dosing for Humulin R. When using a sliding scale, your doctor works with you to create a chart of insulin doses. The doses vary based on how your body responds to insulin and how physically active you are. The doses also vary depending on the amount of carbohydrates you consume in an average day.
To figure out your insulin dose, you first test your blood sugar level. You then find the matching blood sugar level on your sliding scale chart. Next, you match up that blood sugar level with the corresponding insulin dose on the chart.

Pediatric dosage
The manufacturer of Humulin R hasn’t given specific dosage recommendations for children using the drug. Instead, your child’s doctor will recommend a starting dosage. What if I miss a dose?
If you miss a dose of Humulin R, take it as soon as you remember. But if it’s nearly time for your next dose, just skip the missed dose and wait until your next scheduled dose. Don’t take more than one dose of Humulin R at a time. Doing so can increase your risk for serious side effects, such as hypoglycemia (low blood sugar level).
To help make sure that you don’t miss a dose, try setting a reminder on your phone. A medication timer may be useful, too.
Will I need to use this drug long term?
Humulin R is meant to be used as a long-term treatment. If you and your doctor determine that Humulin R is safe and effective for you, you’ll likely take it long term.

Novolin R is also available under the ReliOn brand, which is only available at Wal-Mart stores. The ReliOn form of Novolin R generally costs less than brand-name Novolin R costs.

How insulin affects blood sugar
Normally, after you eat food, it gets broken down in your stomach. And glucose (sugar) from the food travels into your bloodstream. In response to this, your body releases insulin.
Insulin is a hormone that helps move glucose out of your bloodstream and into your body’s cells. Once inside your cells, the glucose can be turned into energy.
Insulin is usually made by your pancreas. But in people with type 1 diabetes, their pancreas isn’t able to make insulin. And in people with type 2 diabetes, their body may still be making insulin, but their cells are resistant to the hormone. (With insulin resistance, your body stops responding to insulin the way it normally should.) Over time, it’s possible that people with type 2 diabetes might not be able to make their own insulin.
If your body is either not making insulin or not responding to it, your blood sugar level can get too high. (A high blood sugar level is called hyperglycemia.) Having high blood sugar levels can damage your organs. And with high blood sugar levels, your cells might not be getting the glucose they need for energy.
What Humulin R does
The active drug in Humulin R, called insulin human, is a copy of the insulin that’s naturally made by your pancreas. This drug is made using recombinant DNA technology. (With this technology, genetic material is combined in order to make the insulin.) Because Humulin R is a copy of natural insulin, your body uses the drug the exact same way it uses natural insulin.
Insulin human works by moving glucose out of your bloodstream and into your cells. This lowers your blood sugar level.
How long does it take to work?
Humulin R has an onset of about 30 minutes. (The term “onset” describes when a drug begins working in your body.) This means it takes Humulin R around 30 minutes to start working after you inject your dose.
Humulin R peak effect
Humulin R reaches its peak (highest) effect in your body around 3 hours after you inject the drug.
Humulin R duration
Humulin R has a duration of action of about 8 hours. (The term “duration of action” describes how long a drug continues working in your body after you take it.) This means that Humulin R keeps working in your body for a total of about 8 hours after you inject your dose. However, Humulin R works most effectively for about 3 to 6 hours after you take it.
Humulin R can cause mild or serious side effects. The following lists contain some of the key side effects that may occur while taking Humulin R. These lists do not include all possible side effects.

Serious side effects and their symptoms can include:
- Hypokalemia (low potassium level in your blood). Symptoms can include:
- constipation
- fatigue (lack of energy)
- muscle cramps
- weakness
- Allergic reaction.*
- Hypoglycemia (low blood sugar level).*

You may wonder how often certain side effects occur with this drug. Here’s some detail on several of the side effects this drug may cause.
Allergic reaction
As with most drugs, some people can have an allergic reaction after taking Humulin R. Symptoms of a mild allergic reaction can include:
- skin rash
- itchiness
- flushing (warmth and redness in your skin)
A more severe allergic reaction is rare but possible. Symptoms of a severe allergic reaction can include:
- swelling under your skin, typically in your eyelids, lips, hands, or feet
- swelling of your tongue, mouth, or throat
- trouble breathing
Call your doctor right away if you have a severe allergic reaction to Humulin R. Call 911 if your symptoms feel life threatening or if you think you’re having a medical emergency.
Weight gain
Weight gain is a common side effect of all insulin products, including Humulin R.
In one clinical study, people with type 2 diabetes who started insulin therapy gained an average of about 4 pounds in their first year of treatment.
Because of how insulin works, this isn’t an unexpected side effect of the drug. Insulin helps your body to move sugar out of your bloodstream and into your cells. Once inside your cells, the sugar is turned into energy. But if the sugar isn’t needed right away for energy, your cells will store it for later use. Excessive sugar is stored as body fat.
Weight gain is also a common symptom in people with diabetes. In fact, people with type 1 diabetes are at higher risk for having excess body weight or obesity than are people who don’t have diabetes.
If you’re concerned about gaining weight while you’re using Humulin R, talk with your doctor. They can help you manage a body weight that’s healthy for you.
Hypoglycemia
You may have hypoglycemia (low blood sugar level) while you’re taking Humulin R. In fact, it’s the most common side effect of the drug.
In one
Talk with your doctor about how often you should check your blood sugar level. Monitoring this level can help prevent hypoglycemia while you and your doctor learn how your body responds to Humulin R.
While it’s important to use insulin to keep your blood sugar from getting too high, having low blood sugar can also be dangerous. Severe hypoglycemia can lead to seizures and even death in some cases.
Certain things can increase your risk of hypoglycemia, such as taking high doses of insulin or using more than one diabetes medication at time. Taking certain other medications that aren’t used for diabetes can also increase the risk of hypoglycemia. Other factors that may increase your risk of hypoglycemia include your diet and exercise habits.
Symptoms of hypoglycemia can include:
- confusion
- drowsiness
- fatigue (lack of energy)
- hunger
- feeling irritable or anxious
- being shaky
Symptoms of hypoglycemia can occur suddenly. And they can quickly become serious if not treated. Be sure to talk with your doctor about monitoring for hypoglycemia while you’re taking Humulin R. And also talk with them about what you should do if you’re having hypoglycemia.

Lipodystrophy
You may experience lipodystrophy while you’re taking Humulin R. Lipodystrophy is a depression in your skin or a thickening of your skin.
Humulin R is given by subcutaneous injection (an injection under your skin). Repeatedly giving subcutaneous injections in the same spots may cause lipodystrophy at or around your injection sites. For this reason, you should alternate the sites where you inject Humulin R. For example, if you inject the drug into your belly one day, you could inject into your thigh or upper arm the next day.
If you’re concerned about developing lipodystrophy while using Humulin R, talk with your doctor. They can recommend ways to help lower your risk of skin effects while you’re using this drug.
Injection site reactions
It’s possible to have injection site reactions at the site where you inject Humulin R. Examples of symptoms that may occur with injection site reactions include:
- itchiness
- pain
- redness or swelling
- skin discoloration
- warmth
You shouldn’t inject Humulin R into an area of your skin that is itchy, red, or swollen. Doing so may increase your risk of a serious reaction, such as bleeding or hematoma (a collection of blood underneath your skin).
If you’re concerned about having injection site reactions while you’re taking Humulin R, talk with your doctor. They can discuss ways to minimize this side effect. And if you have injection site reactions that are bothersome, your doctor may switch you to a different diabetes drug.
When you get Humulin R from the pharmacy, the pharmacist will add an expiration date to the label on either the box of insulin pens or vials of insulin. This date is typically 1 year from the date they dispensed the medication.

Storage
How long a medication remains good can depend on many factors, including how and where you store the medication.
But regardless of where it’s stored, be sure to protect Humulin R from heat and light. Also, avoid freezing Humulin R.
Storing Humulin R before opening
